Multifactorial Optimization of a Molecular Biology Process with LabGenius

LabGenius, creators of the EVA autonomous AI-driven protein engineering platform, faced a recurring bottleneck: optimising single‑stranded DNA (ssDNA) extension reactions for each new library fragment to preserve maximum sequence diversity. The complexity and scale of multifactorial experimental designs made manual re‑optimisation impractical. To address this, LabGenius integrated Synthace’s Antha software into their workflow to drive and automate DoE‑based experimental planning and liquid‑handling execution.

Synthace’s Antha was used to interpret EVA’s DoE outputs, plan multifactorial experiments and autonomously run them on liquid handlers, dramatically reducing effort and consumables. The combined solution delivered up to ~70% time savings (up to 15 hours per full library optimisation and ~5 hours per optimisation iteration), ~35% fewer liquid‑handling actions (≈6 hours saved per run), up to 30% reduction in consumable use, and enabled a second‑iteration design where >81% of conditions exceeded the dsDNA recovery target (25–30 a.u.), ensuring robust, high‑diversity library generation.
