Multifactorial optimisation of a lentiviral vector transfection/transduction system for improved vector titre and robustness

Oxford Biomedica, a world leader in lentiviral vector research and manufacturing, faced the challenge of improving the yield and robustness of its transfection/transduction process for lentiviral vector production. To tackle a complex, multifactorial optimisation that would be hard to run manually, they partnered with Synthace and deployed Synthace’s Antha platform to provide scalable, flexible automation and in silico protocol design for their DoE workflows.

Using Synthace’s Antha DoE suite to drive automated execution (on a Gilson PIPETMAX®), Oxford Biomedica ran larger, more complex experimental designs quickly and reproducibly. The Antha-driven campaign delivered a 3–10× increase in transduction titre, an 81% reduction in pure error (improved robustness), and substantial efficiency gains — including up to ~83% overall time savings, faster setup/execution (physical iterations completed in two weeks), and roughly a 32% reduction in pipette-tip use — demonstrating clear, measurable impact from Synthace’s solution.
