How Oxford Biomedica used DOE to Achieve Improved Titre and Robustness

Oxford Biomedica, a world leader in lentiviral vector research and manufacture, faced a complex multifactorial challenge: optimizing transfection reagent mixes to increase active vector particle production and process robustness. Manual Design of Experiments (DOE) approaches were prohibitively slow and limited in scale, so they partnered with Synthace and used the Synthace platform to tackle the high-dimensional optimization problem.

Using the Synthace platform to plan, automate and execute a reduced two-iteration DOE strategy, Oxford Biomedica screened up to 10 factors with 90–150 runs in a single iteration, enabling rapid workflow setup and smart liquid handling. Synthace-driven automation cut total execution from 49 to 8 hours, delivered up to an 83% time savings across design/planning/execution and a 32% resource saving, and produced a 3–10× increase in vector titre with an 81% reduction in pure error—achieving the objectives in two weeks instead of the typical 12.
