Construct Assembly for the Development of New Therapeutics with Ipsen

IPSEN’s Neuroscience group needed to increase the throughput, flexibility, and efficiency of its Type IIS construct assembly workflow to accelerate discovery of targeted secretion inhibitors (TSIs). To address a labor‑intensive, error‑prone manual cloning process and the difficulty of reprogramming robotics, IPSEN partnered with Synthace and deployed Synthace’s software platform, Antha, to simplify experimental design, simulate protocols in silico, and drive automated liquid‑handling execution.

Using Synthace’s Antha, IPSEN automated Type IIS construct assembly, bacterial transformation, and plating in a device‑agnostic workflow (transferred between Gilson PIPETMAX® and Tecan Freedom EVO®). The team produced 24 constructs in about an hour, cut the time to generate and validate 24–48 constructs from 30 days to 6 days (5× faster), generated 90 constructs (79 expressed), and reused core DNA parts to reduce DNA synthesis costs roughly 10‑fold—increasing walkaway time, throughput, and the number of molecules entering screening to accelerate therapeutic development.
